Determination of organic acids in propionic acid fermentation broth by reversed phase high performance liquid chromatography

Abstract

Water altanlis with dC18 column and mobile phase contained 10 mmol/L potassium dihydrogen phosphate buffer (pH 2. 8) and acetonitrile (95 : 5, V/V) was used to determin organic acid in propioic acid fermenlation system. The flow rate was 1. 00 mL/min and the column temperature was 25°C. Oxalic acid, formic acid, lactic acid, acetic acid, succinic acid and propionic acid were completely separated and determined with UV detector at 215 nm in 9 min. All the correlation coefficients (r) were between 0. 9992 and 1. 0000, and the recovery ranged from 99% to 102%. The relative standard deviation was in the range of 0. 6% -1. 7%. By this method, propionic acid and other organic acids could be rapidly and effectively determined.
