Abstract
The bioconversion of phenylpyruvate to L-phenylalanine using aspartate transaminase in an aqueous two-phase system (ATPS) was investigated. The optimal composition of an aqueous two-phase system was 20% of PEG4000 with 20% of Na2HPO4. Partition coefficients of phenylpyruvate, aspartate transaminase and L-phenylalanine respectively were 8.03, 31.7 and 0.74 in ATPS. The enzymatic yield of L-phenylalanine in ATPS was 78.1%, increased 27.4% higher than in aqueous system when the substrate concentration was 43.4 g/L.
